Description

Currently my focus of study is RNA editing in Trypanosomes, especially newly discovered MRB1 complex  which is involved in guide RNA processing and then the processed guide RNA are send to Editosome for m RNA editing. MRB1 complex has many RNA bindind proteins, like MRB8170,MRB4160,GAP1 and GAP2 , we are interested in knowing the RNA species which they bind to and the strucutre of these proteins. For this we will apply ICLIP and biophysical tecniques like X-ray crystallography and NMR for elucidating the structure.

1# Studying the components of malaria parasite newly discovered

Protein secretion machinery known as PTEX translocon assembly. (AUG10—

STILL ONGOING)

 

Under the guidance of Dr.Amit Sharma, Group leader, Structural and

Computational Biology Group (ICGEB, New Delhi, India).

 

Project based primarily on X-RAY CRYSTALLOGRAPHY based protein

Structure determination of protein assembly.

 

PTEX translocon assembly contains PETEX150, PTEX88, HSP101, EXP2

And TRX2 as protein partners.

 

Trx2 is being crystallized and structure is solved other components of

translocon assembly is now being looked into.

 

We are also looking for other methods for structure determination using

Cryo EM, NMR for structural as well as binding studies and CD for secondary

Structure determination.

 

Confocal studies were also conducted and still going on for other components

along with several assays for protein-protein interactions studies etc.

 

2# Screening of traditional Indian spices for Inhibitory activity of

Acetyl cholinesterase and Butrylcholinesterase Enzyme. (JAN10-MAY10)

Under the Guidance of Dr.Suresh Kumar (BACHELOR'S PROJECT)

The project was mainly based on the ELLMAN based ENZYME inhibitory assays.

 

SKILLS LEARNED

Molecular biology techniques: Cloning, Transformation, PCR etc.

 

Protein purification: Using various techniques like affinity chromatography

Ion exchange chromatography, gpc, Protein identification using Western blotting etc.

 

Crystallization work: setting up the crystallization trays using mosquito, Setting up the

Manual crystallization trays, mounting of crystals, soaking with heavy atoms etc.

 

Biochemical assays: ELISA, Protein-interaction studies using Isothermalcalorimetry (ITC),

Pull down Experiments, Coimmunoprecipitation, and Culturing of Plasmodium Falciparum.

 

Bio-informatics: Using various software’s, Linux language (Ubuntu),

Structure building and refinement programs like coot, pymol etc.
